The aim of the game is to align a given number of pieces of the same colour in a row, column or diagonal. A joker piece can always be substituted for any colour piece. Aligning a given number of pieces will remove these pieces from the board giving you some points. How many pieces you have to remove depends on the level/configuration. But default is five.
To align pieces you can move any piece from its original location to any other location on the board provided that there is a direct path between start and end point of your move. Direct path means that the piece will travel always one step horizontal or vertical, but not diagonal.
Every move you make which will not result in a removal of some pieces will put more pieces on the board. How many depend on the level/configuration but default is three. In this way the board will clutter more and more. You have lost the game if the board is completely filled.
As the game supports arbitrary themes whereever I speak of aligning coloured pieces could also mean aligning pieces of the same shape. In the default theme it is colours though. The joker piece is usually clearly distinguished by color or shape or animation from the normal pieces. In the default theme set the joker piece is a piece which contains all colours.
